چکیده

RecA protein (product of the recA gene), without the help of any other protein, is able in an ATP-dependent reaction to mediate the entire recombination process including the search for homology, homologous pairing and strand transfer (for a review see Radding, 1982; Howard-Flanders et al., 1984b). In this short review, we describe RecA-DNA complexes and how formation of a RecA complex with single-stranded DNA leads to a complete strandexchange reaction between double-stranded linear DNA molecule and homologous to it single-stranded circular DNA molecule.

The synapsis event in the homologous pairing of DNAs: RecA recognizes and pairs less than one helical repeat of DNA.

A key step in homologous recombination is the alignment and pairing of homologous DNAs. The Escherichia coli RecA protein initiates pairing by binding to single-strand DNA, forming a helical nucleoprotein filament. We demonstrate that in the presence of the nonhydrolyzable ATP analogue adenosine 5'-[gamma-thio]triphosphate and ADP, RecA can pair a homologous oligonucleotide 15 bases long with a...

Structure of the hDmc1-ssDNA Filament Reveals the Principles of Its Architecture

In eukaryotes, meiotic recombination is a major source of genetic diversity, but its defects in humans lead to abnormalities such as Down's, Klinefelter's and other syndromes. Human Dmc1 (hDmc1), a RecA/Rad51 homologue, is a recombinase that plays a crucial role in faithful chromosome segregation during meiosis. The initial step of homologous recombination occurs when hDmc1 forms a filament on ...

The meiosis-specific recombinase hDmc1 forms ring structures and interacts with hRad51.

Eukaryotic cells encode two homologs of Escherichia coli RecA protein, Rad51 and Dmc1, which are required for meiotic recombination. Rad51, like E.coli RecA, forms helical nucleoprotein filaments that promote joint molecule and heteroduplex DNA formation. Electron microscopy reveals that the human meiosis-specific recombinase Dmc1 forms ring structures that bind single-stranded (ss) and double-...

Bacillus subtilis RecO and SsbA are crucial for RecA-mediated recombinational DNA repair

Genetic data have revealed that the absence of Bacillus subtilis RecO and one of the end-processing avenues (AddAB or RecJ) renders cells as sensitive to DNA damaging agents as the null recA, suggesting that both end-resection pathways require RecO for recombination. RecA, in the rATP·Mg(2+) bound form (RecA·ATP), is inactive to catalyze DNA recombination between linear double-stranded (ds) DNA...

RecA Protein

The RecA protein of Escherichia coli is the prototypic deoxyribonucleic acid (DNA) strand exchange protein. It assembles on single-stranded DNA to form a helical nucleoprotein filament that is the active species for all RecA protein-dependent functions. This protein– DNA complex is responsible for three mutually exclusive functions: DNA recombination, induction of the DNA-damage SOS response an...
